This progression would be the one most logically followed in an idealized investigation of structure-function relationships. Inasmuch as the methods of protein chemistry are still in a state of rapid evolution, decisions about what to include and not to include were made not only on the basis of the demonstrated value of a method but from an estimation of its potential as well.
